Solutions

Expert Solution

HETP =L/N

High equivalent for theoritical plate

L= length of column

N= number of the theritical plate

TP is directly proprtional to length of column

Solute distributed between st. phase and mobile phase(equilibrium)

When mixture of component is taken into column and run between st. phase and mobilephase, adsorption in stationary than desorbs in mob phase than again adsorb new st. phase.

The process of adsorption and desorption than again adsobs new st. phase this is called a plate.

Plate is an imaginary word, when we take column height high, plate will be more and ads, desoption process tkean very long therefore separation of the mixture very clearly and elution will be purely.

St. phase has no spaced between particles, it is packed perfactly if space there than after running mobile phase due to air bubble,irreagular separation will be take place.

Number of theoritical plate more,separation will be cleared, peak will be separated more, therefore column used as a long as possible, increases separation, narrow peak willbe obtained.

Packing material also used highly uniform, separation willbe increased.

elect all statements below that apply to Column Chromatography. Column chromatography can be used to purify small (< 0.1 g) sample mixtures. The compound in a mixture with the lowest Rf value will exit the column first. Column chromatography is often used to purify large (10 g) sample mixtures. Column chromatography is used mainly as an analytical tool.

Discuss why different elution solvents are often used in the column chromatography. In what order and why should the elution solvents be used? In this experiment would the separation happen if only 10:90 ethyl acetate/hexanes was used as an eluting solvent? Would the separation happen if 20:80 ethyl acetate/hexanes was used first followed by 10:90 ethyl acetate/hexanes? I need at least a paragraph response to this!
