Through Next-Generation Sequencing, we have learned a lot about the human gut microbiome, including community demographics, establishment of the microbiome and microbiome stability. Design a study to determine the effects of daily kombucha consumption on gut microbiome composition and function. Be sure to describe the model system, controls, types of data to be collected, methods of data analysis, and limitations of the study. Also indicate whether the data you gather will provide correlative or causative evidence of the effects of kombucha on the gut microbiome.
Kombucha is a sweet bevarage, fermented and slightly alcoholic. This is a probiotic preparation helps to improve the natural microflora of the gut. It is a live stock contains many microbes like lactobacillus etc.
Next generation sequencing is a advanced system of sequencing contains various methods that fastens and increase the fidelity of the DNA sequencing. The experiment design for the present condition we take random sample of people from a community who is same age group with similar dietary habits and other lifestyle. A set of healthy individuals with almost similar probable gut health could be considered and carried a specimens under study. Half of them would be given with a standard dose of kumbucha and other are given with a same amount of water daily. People given with water are considered as controls or placibo . Taking equal number of controls helps us to elliviate interfearence if data due to individual differences. The DNAsamples from gut sufaces are collected from all of them sequentially and community DNA of the gut microflora have collected. The DNA samples are amplified in PCR and subjected to NGS to understand their meta genomic's. Here the number of bacterial species in the each sample, and their respective amounts (approximate value), and the composition at early and delayed phase of kumbucha consumption are evaluated. The health of gut considered to be increased by improving gut microflora in individuals who has taken daily kumbucha and compared with controls. Here the change invariety and amount of gut bacteria indicated the gut health. The data analysed and tested for its significance using chisquare test. The null hypotheses cold be similar to ' there is no significant difference in in the beneficial microflora of gut in samples compared with controls. And the values if shows any significant difference than it could be considered proved that the daily kumbucha consumption improves the gut health
