In in vitro transcription: Is the RNA produced sense or antisense? How can you tell? How could you produce the opposite strand
In in vitro transcription RNA produced can be any of them sense or antisense, it depends on the template we are transcripting. If 5' end of coding strand is downstream the promoter then it will produce sense RNA and if 5' end of noncoding strand is downstream of +1 G it will produce antisense RNA.
If we want to produce the opposite strand, we will have to use a vector which has promoter and adjacent to it a restriction site, now we will join it to our insert such that 5' end of opposite strand ligate downstream of promoter. Now we will linearize the plasmid and RNA transcribed will be of opposite strand.
