Transferring has been shown to be one of the most efficient cellular receptor molecule for targeted drug delivery system in the brain. Studies have shown that the ligand molecules which bind to this receptor can be modified using various physico-chemical processes to enhance their ability to cross the blood-brain-barrier and improve their entry into the brain. Following methods can be used to perform this task:

  1. Chemical modifications: The target ligand can be modified by linking it to chemical linkers such as maleimide linkers etc.
  2. Biological methods: The biological solubility of the ligand can be enhanced by increasing their solubility in their solubility by addition of specific chemicals such as liposome, polyethylene glycol, glutaraldehyde etc.
  3. Entrapment: The target molecule/ligand can be entrapped into non-reactive polymers such as liposomes to enhance their solubility.
  4. Receptor-modification: The receptor-mediated binding of the ligand can be enhanced by binding of the molecule biological modulators of receptor binding such as avidin-biotin complex to enhance their binding.

Thus, these are some of the methodologies to enhance the receptor mediated ligand binding in brain.
