It is described that for a human protein that is expressed only in the brain, There is an identical but smaller variant that is expressed only in the liver. What experiments would you use to determine the origin of this differential expression?

Expert Solution

- Yes, we all know brain and liver both are complex organ that controls a variety of bodily fuctions. Brain : It perfomed fuction like maintenance of homeostasis, processing of sensory information and genration of behaviour.Liver : production of bile ,hormone and vitamins, storage of glycogen and removal of toxic substsnce.

The human genome projects revealed the gentic code ,enabling extensive analysis of gene expression in tissue and organ samples in the context of evolution, physiology and disease. The transcriptomes of 26 pheripheral human organs and three fronatal cortex samples were analyzed using next generation sequencing based on specimens. The transcriptome of each sample was quantified using RNAseq to determine the normalized mRNA abundance.High correlation between biological replicates indicates low inter- individual variability in gene expression.

Transcriptome analysis of the liver can be visualized with regard to specificity and distribution of transcribed mRNA molecules. Specificity illustrate teh number of gene with elevated or non elevated expression in the liver compared to other tissues. In depth analysis of elevated genes in liver using antibody based protein profiling allowed us to visualize the expression patterns of these proteins in different fuctional compartments including plasma proteins, enzymes, bile proteins and transporters.

The protein REST is a transcription factor that repressesthe expression of neuronal genes (genes only expressed in neurons). Predict whether REST would be found bound to the promoter of each gene in the designated tissue (think about whether they are expressed in each tissue). Barnase is a bacterial protein that is lethal to the cell unless it is expressed with its inhibitor barstar. Barstar must bind to barnase very quickly before barnase kills the cell that made it. Therefore, barnase and barstar use electrostatic attraction to find each other quickly (Schreiber and Fersht, 1996). a) At neutral pH, barnase has a charge +1e and barstar has a charge −6e. You are trying to design a protein that will be expressed in Escherichia coli and secreted outside of the cell for purification and use as a pharmaceutical. E. coli is a gram-negative cell and the protein folds after it has exited the cell. Which secretion system would work best for this project? Support your answer with evidence based on the properties of E. coli, the protein and the secretion system.

You’re interested in studying the catalytic activity of a rare protein expressed in an exotic plant but present in very low amounts in plant tissues. Your supervisor asks you to search an NCBI genomic database for a potential homologous DNA sequence and you think you found a good match. Propose a strategy to obtain large quantities of this plant protein for your studies. Your complete experimental strategy must be outlined to receive full marks for this question.
