If I mutated exon 6 of the BACE1 gene such that the BACE1 lncRNA would not bind the BACE1 mRNA, what affect would it have on the accumulation of beta-amyloid plaques?
INTRODUCTION
BACE1:
(1) It is the enzyme found in human and is
encoded by BACE1 gene
(2) It is important in formation of myelin sheath
in peripheral nerve cells.
Inc RNA-:it is long non coding RNA defined as being transcripts with length exceeding 200 nucleotides that are not translated in protein.
So the question is to know the affect on accumalation of beta amyloid plaque.If there is mutation in the amyloid precursor protein(APP) gene cause early onset of autosomal dominant Alzheimers disease.
And if we read the pathology of Alzheimers disease,there is gross atrophy of cerebral cortex associated with secondary enlargement of ventricular system.And the microscopy revealed following
1.Extracellular neuritic plaque containing A beta amyloid.
2. Silver staining neurofibrillary tangles in cytoplasm that represent abnormally phosphorylated tau protein.
3. Accumulation of A beta amyloid in arterioles(amyloid angiopathy).
GENETICS:- Abeta 40 and Abeta 42 are the main component of senile plaques in Alzheimers disease brain parenchyma.
