c) Suppose you isolated a mutant form of the Apr2/3 complex that did not require a mother filament to bind to in order to nucleate actin filament assembly. How would you expect the mutant form to behave (that is, what would you expect to observe) in:
1) a TIRF assay of actin filament assembly
2) a migrating cell

Actin-Related Proteins ARP2 and ARP3, the structure of monomeric actin and serve as nucleation sites for new actin filaments. But in mutant ARP2/3 its normal function is disrupted.
1) a TIRF assay of actin filament assembly:- Total internal reflection fluorescence microscope, is mainly used for investigating molecule trafficking and how it occurs e.g. in cytoskeleton assembly.
So, as we know arp2/3 is a major component of the actin cytoskeleton and is found in most actin cytoskeleton-containing eukaryotic cells and helps their assembly. But here arp2/3 prptein in mutated, so in TIRF study proper molecular trafficking in not indicating, also some abnormal results observed. Cellular functions abnormality is also observed.
2) a migrating cell:-
We know that during cell migration cytoskeleton and actin filaments are play a major role . But for the mutated ARP2/3 these all functions are show abnormality. The abnormal regulation also observed, like cell locomotion, phagocytosis, and intracellular motility of lipid vesicles not done properly. Thus It also cause the death of the cells.
