Why is the Leibovitvz’s L-15 medium is important for adherent cell culture? Identify five key components of this medium. Explain why facal bovine serum is added to your medium.

Solutions

Expert Solution

Leibovitz L-15 Medium was specifically designed to grow cells in a carbon dioxide free atmosphere. The standard sodium bicarbonate/ carbon dioxide buffering system is replaced by a combination of free base amino acids, phosphate buffers, and higher levels of galactose and sodium pyruvate. As a result, the medium does not require addition with sodium bicarbonate and can be used under conditions of free gaseous exchange with the atmosphere. The medium can be used to grow human tumor cells and embryonic cells. The medium is frequently used in diagnostic virology where tissue cell lines or strains need to be grown in closed systems. Leibovitz medium avoids to need for frequent medium change.

The facal bovine serum is added to Leibovitvz’s L-15 medium because it is used as a supplement to cell culture media to help sustain cell growth and metabolism. It alsos upports growth of monkey kidney cells and primary explants of embryonic and adult human tissue. This product is for research use only.

Five key components of Leibovitz's L-15 Medium iare phosphates free base amino acids, galactose, L-glutamine, phenol red, HEPES, sodium bicarbonate etc

For cell culture, various laboratories use different volumes of culture medium in their culture flasks according to their conventions. However, many do not consider how this might affect the diffusion time of O2 from the gas/liquid interface to the cells attached to the bottom of the flask. To address this, it is useful to have an understanding of how the O2 diffusion time might be influences by the volume of medium added. A typical cell culture flask has a surface...

Because everyone in the world is using lung cell culture medium, there is a shortage. But, no problem, you can make a homemade version using basic components. The final concentration of the components in culture medium are 130 mM sodium chloride, 2.5 mM potassium chloride, 25 mM sodium bicarbonate, and 100 μg/mL BSA. You need to prepare a 10 x concentrate of culture medium. You have the following stock solutions: 5 M sodium chloride, 0.5 M potassium chloride, 2.5 M...
