The two enzymes whose dysreguation causes accumulation of succinate and fumarate are - Succinyl CoA synthetase and succinate dehydrogenase. Succinyl CoA is converted to succinate by the action of the enzyme Succinyl CoA synthetase and fumarate is formed from succinate by the enzyme succinate dehydrogenease.
If fumrate is not formed, the oxaloacetate formation will also be affected as fumarate is converted to malate by the enzyme fumarase and subsequently malate is converted to oxaloacetate by the enzyme Malate dehydrogenase. If oxaloacetate is not produced, Acetyl Co A , formed after oxidative decarboxylation of pyruvate (The end product of Glycolysis) can not condense with oxaloacetate and TCA cycle will be drastically affected.
