In the DNA extraction from cheek cells, protocol you used cold ethanol to:

A.) Precipitate the DNA B.) Break open the nuclear membrane C.) Separate the nucleus from the other organelles D.) Denature the nuclear proteins

Solutions

Expert Solution

DNA isolation occurs in many steps

Coming to question

In DNA extraction of cheek cells cold ethanol is used as it dehydrate DNA and it become insoluble in water and precipitate and appear as white thread like structure in the tube and can be extracted

Use of chilled ethanol also yield more DNA as it prevent activation of DNAse enzyme which can split DNA which can result in low quantity of DNA extracted

Hence option A precipitate DNA is correct answer other option can be eliminated

DNA EXTRACTION & PURITY PROTOCOL Add a small piece of peeled banana (2-3 cm) and 12mL extraction buffer (NaCl & Sodium Citrate) into the beaker. This buffer contains salts necessary for the precipitation of the DNA. The ions in the solution make it more polar facilitating the dissolving of the DNA. Add 3mL detergent into the beaker. This will break open the cells and dissolve the DNA. Detergent breaks up the lipids of the plasma and nuclear membranes Questions: a....

im the DNA extraction from a banana procedure it states: "Try to prevent the alcohol from mixing with the banana mixture." Subsequent steps also emphasize that you should "avoid any sudden disturbance" Why is this important? Consider what is present in the test tube and whag the purpose of these steps is. why is it important to keep the alcohol and banana mixture from mixing?

a. You start a culture of stem cells and you use a protocol to differentiate them into beta cells. To make sure they are truly beta cells. First you want to check whether the insulin gene is expressed at higher levels in the differentiated vs. the undifferentiated cells. What methods that we studies would you use and describe the step by step procedure. b. Then you want to demonstrate that they secrete insulin when you place glucose in their culture...
