Regarding DNA extraction:
1) Describe the purpose of each of the following steps
or reagents used in DNA isolation (in detail):
- Using fresh vs. dried specimens
- Grinding or tearing tissue
- Lysis solution
- TE buffer
I. Describe one DNA extraction method. In your description
compare and contrast the method you chose with the other DNA
extraction methods. Additionally, describe what types of forensic
samples are better suited for this type of extraction and why.
a)Given the three types of DNA techniques (STR, mtDNA, and Y-STRs)
briefly compare and contrast all of them and in your opinion state
why you believe one to be better than the other. YOU MUST SUPPORT
YOUR ANSWER WITH AN EXAMPLE...
E. coli DNA ligase is different than eukaryotic and
bacteriophage DNA ligase. Describe how the reactions would be
different and any other differences in these enzymes that could
affect their use in the lab? Which ones are used in the lab and for
what?
Explain the process of transcription. Be sure to include (these
are in no particular order): DNA, mRNA, template strand,
non-template strand, antiparallel, 5' and 3', RNA polymerase,
promoter region (TATA Box), elongation, termination, 5' cap, poly-A
tail, where the process takes place, primary RNA transcript versus
finished mRNA strand, exons, and introns.
