First comparison between protodrugs phenytoin and carbazepine

1.Phenytoin

It is Na+ channel blocker

First line management in GTCs ,partial seizures,neuropathic pain,antiarrhythmic drug.

There is increased risk of toxicity in case of overdose.

Side-effects include

Hyperplasia of the gum

Lymphadenopathy

Ataxia, Nystagmus, teratogenic.

2.Carbamazepine;

Na+ channel blocker

It is drug of choice for partial seizures, trigeminal neurolgia, bipolar disorders and acute mania.

Prodrug eslicarbazepine is less toxic

Side effects include

Hyponatremia

Eosinophilia

Aplastic anemia

Diplopia

Splenomegaly.

Second comparison between protodrugs Ethosuximide and lamotrigine

1.Ethosuximide

It is a Ca+2 channel blocker

It is used as drug of choice when it seizures are absent.

Side effects include nause , vomiting, neurotoxicity,bone marrow supression,SLE.

2.Lamotrigine

It is a Ca+2 channel blocker

Uses:in first line GTCs,myoclonic seizures, partial seizures,and in bipolar disorders.

Side effects include; nausea , vomiting ,ataxia ,diplopia.
