synthesis reaction for dopamine to mdma

synthesis reaction for dopamine to mdma

Solutions

Expert Solution

Dopamine first treated with NBS (N-Bromosuccinamide) in CCl4 solvent which selectively brominates benzylic position. This reaction follows free radical mechanism. Then subsequent dehydrohalogenation (elimination) using alkaline condition forms (A).

Benzene 1,2 diol of (A) is protected using formaldehyde in acidic conditions gives (B)

Then (B) on treatment with BuLi gives lithiated compound forming vinylic anion. This vinylic anion is reacted with Methyl bromide gives vinyl methylated product (C)

The limiting reagent is used for methylation of amino group of (C) which otherwise gives excessively methylated product i.e. quaternary ammonium salt. This gives (D) in good yield.

(D) Then hydrogenated with mild hydrogenating agent like Pd in BaSO4 which gives MDMA.
