1. The function of ferrous sulphate in Kligler's iron agar (KIA )medium is used to detect the hydrogen sulfide gas production by producing black precipitate with sodium thiosulfates reduction . It is found either as ring formation near top of the butt or throughout the butt.
2. Gelatin hydrolysis test is used to test the secretion of enzyme gelatinase. When a tube of nutrient gelatin is stabbed with gelatinase positive organism , liquefaction of medium occurs within 7 days. If the tube shows negative even after 7 days, additional 7 days are incubated to finish the test.so approximately 14 days are needed to finish the gelatin hydrolysis test.
3. The inoculum of pure culture is stabbed in tube of nutrient gelatin and incubated at 35-37°c for 24 hrs . To determine whether the gelatin is actually digested by gelatinase enzyme, the incubated tube is placed in the refrigerator for few minutes causing undigested gelatin to resolidify.
