• Autoclave is an instrument which contain pressure chamber to carry out microbiology processes.
• The use of a autoclave is to sterilise media, gloves, glass, forceps, needles, pipette tips etc.
• It is a moist sterilisation technique to sterilise mainly the things use under biosafety cabinet.
• Normal pressure and temperature of a working autoclave is about 121•C and 15 Ibs pressure.
• Working of autoclave: Steam entered in the autoclave passes through the operating valve to enter inside rear of chamber which is behind the baffle plate. pressure regulator is present to control the pressure around 15 Ibs and safety valve control the over pressure. Pressure is to raise and reach the temperature at 121•C.

Major need for an autoclave

1) Sterile equipments using in Laminar air flow should not get contaminate

2) Using an autoclaved product avoid false positive results.

3) Autoclave is to kill microbes using high pressure and temperature to get accurate and stable results in the experiments.

In microbiology or infectious disease world the chances of contamination and cross contamination are excess. Autoclave has a major role in controlling it.
