Discuss the structure of promoter for RNA Polymerase
II
Ans. The core promoter of RNA polymerase II includes two segments.They are TATA box and Initiator (Inr) sequence. TATA box, also known as -30, is a consensus sequence in promoter of RNA polymerase II and the sequence is 5'-TATAXAX-3' where X = A or T). Location of Initiator (Inr) sequence is around the transcription start site i.e. +1. The consensus sequence of initiator (Inr) sequence is 5'-Py2 CAPy5 -3' and is pyrimidine rich sequence. Besides this core promoter, there are cis-acting promoter sequences. These are regulatory sequences. They act as binding sites for various regulatory proteins thereby controlling the expression of gene. Generally, their location is at upstream side of TATA box but sometimes they are also present downstream in some genes. CAAT box and GC box are two cis-acting regulatory elements in numerous eukaryotic genes. NF-1 and NF-Y are activators and recognize CAAT box. Another activator Sp1 recognizes GC box. Some genes possess Downstream Promoter Element (DPE) and their position is at +28 to +32. The sequences in DPE are variable in different genes.
