What would be the effect of removing the ligand binding domain of Estrogen receptor and replacing it with the ligand binding domain of retinoic acid receptor? (5 Points)
What would be the effect of mutating the DNA binding domain of the glucocorticoid receptor? (5 Points)
Ans :- Estrogens pass directly into cells throughout the body, so the cell can use receptors, that are in the nucleus, right at the site of action on the DNA.When estrogen enters the nucleus, it binds to the estrogen receptor, causing it to pair up and form a dimer.
Glucocorticoid receptor (GR, or GCR) also known as NR3C1(nuclear receptor subfamily 3, group C, member 1) is the receptor to which cortisol and other glucocorticoids bind.
The GR is expressed in almost every cell in the body and regulates genes controlling the development, metabolism, and immune response.
The glucocorticoid receptor (GR) is a ubiquitously expressed DNA-binding transcription factor (TF) that directly regulates thousands of genes associated with stress response, inflammation, and apoptosis, if mutation occur in them it will lead to responses of body in conditions in stress and inflammation is deprived.
