Cannabinoid receptors CB1 and CB2 generally signal through a type of heterotrimeric G protein known as Gi/o ( G-protein coupled receptors). What is the difference between Gαs(that stimulated analytic cyclic) and Gi/o? How are they similar, and how are they different?
ANS : Heterotrimeric G protein, also sometimes referred to as the "large" G proteins (as opposed to the subclass of smaller, monomeric small GTPases) are membrane-associated G proteins that form a heterotrimeric complex. G protein is that heterotrimeric proteins bind to their cell-surface receptors, called G protein-coupled receptors, directly. G proteins are made up of alpha (α), beta (β) and gamma (γ) subunits.here four main families of G proteins: Gi/Go, Gq, Gs, and G12/13.Gi/o work as a Inhibition of adenylate cyclase, opens K+-channels (via β/γ subunits), closes Ca2+-channels in signal transduction .
G-alpha subunit (Gαs) and its role in endosomal receptor trafficking and signaling is well documented .Gαs has been reported to be activated and signal from the intracellular compartment following cell-surface activation and internalization of parathyroid hormone receptor .
