a). Hemoglobin (Hb) contains four heme units (tetramer)and four globular proteins sub-units.
Hb has two histidine group one is DISTAL and another one PROXIMAL. Hence, histidine group (DISTAL) situated near Iron and assisted the binding of dioxygen in "end-on-bent" conformation, this particular bent discourage the binding of carbon monoxide (CO) to haem iron.
If Hb molecules bound to CO then CO-Hb referred to as T-state (Tense) of Hb at low pH.
b) Bisphosphoglycerate (BPG) reduces the affinity of Hb with oxygen it binds between two beta units of Hb where it stabilizes the T-state into R-state (relax) of Hb, at high pH.it also observed that at high altitude (mountains) the amount of BPG increases in blood.it does not affect the affinity of the affinity of Hb with oxygen in lungs but decrease the affinity in tissues.
