Compare and contrast the different types of inhibitors, with examples, including: transition state inhibitors, suicide inhibitors, irreversible inhibitors, competitive and noncompetitive inhibitors
The transition state inhibitars binds to the active site of the enzyme in the enzyme catalysed reaction . As the enzyme 's active site location is occupied by the inhibitors and there are very less enzyme available for the catalystsation reaction. The rate of reaction decreses examples are :- Neuraminidase inhibitor, sequinavir that is used in HIV treatment .
Suicide inhibition is an example of irreversible inhibition in
which the inhibitors takes part in the reaction and convert into
more potent inhibitor and inhibit the cataysis of reaction example
:- asprin in high doses.
Irreversible inhibitors or the non competitive inhibitors are
generally heavy metals oxidising agent . They bind to the enzyme
other than active site and inhibits the process of catalysis .
Increasing the concentration of substrate has no effect on the rate
of catalysis examples are cyanide , fluoride , british anti
lewisite .
Incompetitive inhibition , the inhibitor binds to the active site
of the enzyme and there is a competition between enzyme and
inhibitors for binding with active site of enzyme . The rate of
reaction increases on increasing the substrate concentration
example :- malonate is the inhibitor of succinyl dehydrogenase
.
