Answer:

  • The expression of Trp operon is regulated by transcriptional atteunation.
  • Trp operon contains a leader sequence between the operator and the structural genes.
  • When the tryptophan is low, the structural genes including leader sequence gets transcribed but when tryptophan level is high, structural genes are not transcribed but the short sequence of 140 ntds is transcribed.
  • The leader sequence contains four regions - Region 1, has 2 trp codons, Region 2, and Region 3 and 4 that has poly (U).
  • When tryptophan is high in the medium, Region 1&2 and Region 3&4 base pairs to form hair pin structure, transcription stops.
  • But during its low level, region 2 and 3 base pairs, inhibiting the base pairing between region 3&4 and transcription continues.
  • So if two Trp codons changed to Gly, then the ribosome will continue the translation of leader sequence and will stop at region 2 (contains stop codon) resulting in the Region 3&4 base pairing, turning OFF the transcription.
