The general equation of a rectangular hyperbola is xy=1. Here we take substrate concentration [S] in X-axis and rate of reaction (r) in Y-axis. As xy=1,that means x=1/y

Or,rate of reaction is inversely proportional to substrate concentration. => r 1/[S]

Rate of reaction is inversely proportional to substrate concentration means with increase in substrate concentration rate of reaction decreases.This is only possible in case of negative cooperativity or inhibitory factors are present to disturb the substrate.

Option 1 is not valid because there is no case of no inteaction between binding sites.Option 2 is also not true because Michealis- menten assumption is not valid as it is rectangular hyperbola case given,but the actual graph of Michaelis menten equation is a saturation curve.Option 3 is also incorrect, because one binding site will also follow saturation curve.
