A new tumour suppressor gene (Tu015) has been identified which is associated with breast and ovarian cancer. Describe an experimental strategy using genetically modified mice to confirm that Tu015 is a tumour suppressor gene, rather than an oncogene, responsible for breast and ovarian tumours. Please explain briefly.

Expert Solution

Genetically modified mice models using technology is available to control the overexpression and gene expression i.e done either through out mouse or in specific organ.Same method can be used in case to coform activity tumor suppression gene. With targeted mutagenesis scientists make specific models ,single gene defects can be readily mimicked in mice once associated gene is cloned.Mutation in inhibin gene appear to cause tumours in the ovaries;which shows that it is a potentil tumour suppressor gene.Eulopidy condition in case of Tu015 ,a cell or organisam has one or more than one complete set of chromosomes. Here a mice can be used to check the above condition by genetical modification .Tumor supperssor gene on adding to it there will be effect on their growth and functioning of cells ,whose loss of function leads to tumerogeneisis, which insights into mechanisam of human cancer. On specifically conducting on specific tissue types we can coform the Tu015 responsible foe breast and ovarian cancer. This cofirmation comes from identification of growth on tissues and loss of heterozygosity which occurs when Tu015 become inactivated ,when tumor suppressor gene mutation acquires.

c. A protein called Merlin (Nf2) has been identified as a tumor suppressor. An explanation for its role as a tumor suppressor is likely found in Merlin’s influence on vesicular trafficking! Researchers labeled a v-SNARE with green florescent protein to monitor vesicular trafficking in cells lacking merlin protein. Why would monitoring a v-SNARE allow researchers to track vesicular trafficking? Rubric (2): explanation shows understanding of where vSNARES are present in the cell. Answer: Once a vesicle is formed with its...

Sterility is associated with loss of function (LOF) at a gene Z. The gene has 5000 bases where LOF mutations can occur. Assuming the human mutation rate is 2x10^-8 per generation. Homozygotes for LOF alleles at Z are sterile, and heterozygotes have mild sterility issues. The heterozygotes for LOF alleles at Z have 20% fewer children than an average individual I the population, but suffer no other ill effects. A) What is the frequency of the Z allele under mutation-selection...

Among the alleles for the gene responsible for cystic fibrosis that have been identified, one mutant has been identified in which three amino acids in the middle of the protein sequence are changed.  What change in the DNA sequence of the gene could cause such a mutation? You studied a small, isolated human population and identified a new gene A. You found these numbers of individuals with these genotypes (capital letter represents dominance): 250 individuals of AA genotype, 500 individuals of Aa phenotypes and only 1 aa genotype, who is a 3 years old boy. Is gene A in Hardy-Weinberg equilibrium? Describe the possible reasons why gene A is (or isn’t) at HardWeinberg equilibrium. What can you conclude from the collected data?

The human gene for a protein hormone has been isolated. Scientists want to introduce this gene into a bacterial host, modifying the bacteria so that it can be used to produce this hormone. After successfully introducing the gene on a plasmid, the bacteria produced the protein hormone, but the protein was defective, longer than normal, with intervening stretches of amino acids not found in the native protein. It has been noted that breast cancer tumor biopsies appear to contain a heterogenous population of cells. a. Describe two ways (briefly) that you can show that there indeed is a heterogenous population of cells b. Do you think the different cells have differing potential to generate tumors and if so how can you definitively show this?
