If a circular plasmid is cut one with a restriction enzyme what is the result?

Cutting of a circular plasmid once with a restriction enzyme would linearize the plasmid, if the plasmid has only one site for the restriction enzyme. If the plasmid has more than one site for restriction enzymes, the linearized plasmid would be fragmented into different pieces. Number of fragments depends on number of restriction site.

How do we determine how many times a restriction enzyme cut?

Agarose electrophoresis of digestion productions, after digestion with restriction enzyme, would give clear idea about number of restriction sites.

For instance, if the plasmid has 2 restriction sites for EcoRI enzyme, it would give 2 fragments (bands on the gel). Cutting at first site would linearize the plasmid, while cutting at the second site would cleave the linearized plasmid into 2 fragments.

If the plasmid has 3 restriction sites for EcoRI enzyme, it would give 3 fragments (bands on the gel)
