Importance of incubation temperature are as given below :-

Incubation temperature affects the growth and affects the fungi in their development of isolation plates. Soil surface , temperatures are buffered by the cooling affrects of the soil water evaporation and the poor heat conductivity of the soil. Isolation schemes of the soil fungi should take into account prevailing soil temperatures and there seasonal amplitudes and localized patterns of shading and isolation.In forrest openings, grasslands, desearts isolation may elevate surface soil temperatures.So many incubators maintain temperature cycle they can survive between the the temperature in which humans can live in, these incubators can survive in heating and cooling cyles inside humans.That means the actual temperature may transiently be as high as 20 and 37 degree celcius.This could be high enough to seriously affect some sensitive bacteria.To avoid this possibility, people aften leave a small margin for error or safety and grow the bacteria at 35C. Slightly cooler or higher can kill those bacteria.

Temperature of incubation is between (30C-35C) range is used because of approx human body temperatures in which mesophilic organisms grow.Many potential microbial contaminants would be of human origin such as the micrococci and staphylococci of skin flora. Microbes can be roughly classified according to the range of temperature at whic they can grow.The growth rates are the highest at the optimum temperature for the organism and the the lowest temperature at which the organism can survive and can replicate themselves at maximum growth temperature.We know that organism can be categorized as mesophiles called middle loving are adapted to moderate temperatures with optimal growth temperatures ranging from room temperatures about 20to 45 dgree celcius.As expected from the core temperatures of the human body 37C, normal human microbiota and the pathoges are mesophiles ex-Salmpnella spp., and Lactobacillus spp.

The following bacteria can grow under following temperatures;-

psychrophiles- between(below 0 to 20C), mesophiles between(15-45C), Thermophiles between(40-80C),Hyperthermophiles between(80-121C).
