Consider cells treated with a drug that enhances the ability of the Trimeric G-protein to associate with GTP

The overall concentration of Ca​​​2+ in the cell will increase of the drug enhance the ability of trimeric G protein to associate with GTP. Because if the GTP remains attached G protein, the complex of G​​​​​​​​​​​q​​​​​-protein will remain activated. So, phospholipase C will continuously act on PIP​​​​2 to break into DAG and IP​​​​​3​. IP3 will remain binds to it's receptors on Endoplasmic reticulum and thus release calcium ions into the cell through Calcium ion release channels. There will be no need of any ligand to bind to GPCR if this drug is given which enhance the ability of G protein to remain associate with GTP.
