simple staining lab 3 simple stain experiment
Ans: Bacterial culture is stained that produces a contrast between culture and the background.
Simple staining procedure-
1. Microscope slides are washed and cleaned thoroughly
2. Slide surface onto which smear has to be spread is sterilized under the flame
3. The inoculating loop is flamed
4. Culture tube cap is removed and kept under the thumb
5. Incoluating loop is shown under the flame and the loop is allowed to cool down for a minute
6. Culture is picked up using a loop head and dropped onto the agar plate without digging the plate
7. Culture is spread over the plate
8. The bacterial smear is dried by showing the underside of the slide under the flame
9. Culture is stained using staining solutions and allowing it to remain covered for below mentioned time-
Methylene blue - 1 min
Crystal violet - 30 seconds
Carbol fuchsin - 20 seconds
10. Excess staining solution is rinsed off using tap water at the end of the incubation period
11. A stained smear is placed under the microscope
