Electron transport chain is the process of transfer of electrons from one electron carrier to the other. All the electron carriers are arranged in increasing order of their electrode potential. Electrode potential define the affinity of a protein molecule towards electron. Higher the affinity, higher the value of electrode potential. This process take place in inner mitochondrial membrane.
So if we want to isolate electron carriers, we should use inner mitochondrial membrane. Because the proteins are transmembrane proteins, the complete structure of inner mitochondrial membrane needs to be disrupted. This can be done by using detergents.
I will use heart of cow for isolating mitochondria. It is because heart is the most active organ of any animal. It pumps constantly. The day it will stop pumping, we will die. So if it is pumping constantly, it needs a constant amount of energy for that. That will be generated only by oxidative phosphorylation in mitochondria and therefore this organ will have the maximum number of mitochondria.
