Differential centrifugation creates a sort of gradient where in denser heavy particles settle at bottom and lighter counter parts stay in supernatent .... since there is difference in (S) swedberg unit for prokaryotic and eukaryotic subunits as prokaryotic ribosomes are 70s and eukaryotic ribosomes are 80s so they stop sedimenting at respective sedimentation coffecient in the gradient. Hence can be seperated thereof
