1. List the arrangement of serum proteins from anode to cathode following agarose gel electrophoresis.

2. State one cause of each the following problems: no migration of proteins, tailing of bands, thin bands, “blobby” patterns.

Solutions

Expert Solution

Answer to question number 1:

Arrangement of serum proteins from anode to cathode following agarose gel electrophoresis:

Albumin

Alpha1  (Alpha1antitrypsin, globulin, transcortin)

Alpha2  ( Ceruloplasmin, Alpha2 macroglobulin, haptoglobin)

Beta1 (Transferrin)

Beta2 ( Beta-lipoprotein)

Gamma fraction(Immunoglobulin)

Answer to question number 2:

One cause of each of the following problems are:

  • No migration of proteins:  When the pH of stacking and resolving gel buffers is not correct
  • Tailing of bands: When the agarose gel is disturbed during the process of solidification it leads to non uniform density along the gel. As a result the bands may appear curved, wavy, or twisted.
  • Thin bands:  Very less amount of DNA is loaded in the gel. Gel percentage that is not suitable for the band size to get better resolution.
  • “blobby” patterns: The voltage at which we run our gel because uneven heat transfer can also result in patterning bands or blobby patterns.

Linda has asked Brenn to prepare a 1% agarose gel for a gel electrophoresis analysis. But before he could do that, he needs to prepare 600 ml of 1x TAE from 95x TAE. How much 95x TAE and water should he add to create the 1x TAE solution? Afterwards, how much agarose does he need to weigh out, in grams, and add into 50 ml of 1x TAE?
