The following methods can be used to isolate primary cell lines from tissue :
A. Mechanical disaggregation :
This technique is generally used for the disaggregation of soft tissues e.g. spleen, brain, soft tumors etc. It involves chopping or slicing of tissue followed by sieving, syringing etc. to collect the cells.
B. Enzymatic digestion :
This technique is generally employed when high recovery of cells is required from a tissue of interest. Enzymes e.g. trypsin, collagenase etc. are used to perform this technique.
C. Explat culture :
Primary explant culture technique is performed when cells have to be isolated by disaggregating small quantities of tissues. Tissues in basal salt solution is chopped, washed and incubated for a period by adding appropriate medium. Then primary explant are collected and used for further experiments.
