  • The ATP synthesis process occurs in mitochondria coupled with proton gradient. As the synthesis of a molecule of ATP requires 50 KJ thus is combined with proton gradient.
  • Proton is pumped from the matrix to of inner mitochondria that are transported in relation to electron transport from NADH to O2 in the electron transport chain. Such that the energy released during electron transport to O2 from NADH is used to pump proton to the lumen.
  • Thus the electrons are transported occurs in the matrix so that it is electronegative and the protons are pumped to the lumen so that the lumen is electropositive put the H+ pumped inside equals to the electrons pumped out so electrical they are neutral.
  • And the protons pumped inside are made to pump out through small pores in F0 complex as fast as proton was pumped into by electron motive force. And the energy generated through this process is used to generate ATP so that the net energy requirement becomes almost zero.
  • And the F1 complex, which is having an enzymatic activity as it has ATPase cause the hydrolysis reaction that is ADP + Pi ATP + H2O
  • The proton gradient is coupled to ATP synthesis more clearly the substrate oxidation and phosphorylation are coupled that is while substrate, undergoes oxidation electrons are transported to it and with the electron, transport protons are pumped to the inner membrane and this energy is used to generate ATP by phosphorylation of ADP.
