Expression vectors ______.

a. require a promotor if used in yeast but not in bacteria.

b. are created by oligonucleotide-directed mutagenesis.

c. are only used in bacteria and yeast.

d. can allow for production of fusion proteins.

Solutions

Expert Solution

Answer: d. can allow for production of fusion proteins.

Expression vectors must have expression signals such as strong promoters and are used in bacteria, yeast, baculovirus, plant, mammalian and cell-free systems (prokaryotes & eukaryotes as well). These are used for site-directed mutagenesis. prokaryotes expression vectors would have a Shine-Dalgarno sequence, while eukaryotes expression vectors would contain the Kozak consensus sequence. vectors are generally designed for cloning a foreign gene into a host genome to produce proteins which are not produced by host. Fusion of one gene to another gene will produce a fusion protein. For example pGEX-TT expression vector used for production of GST fusion proteins, it Contains the polylinker from pEG-KT, tac promoter, lacIq repressor, ampicillin resistance.

Bacteria and other kinds of pathogens (like the yeast Candida auris) used to be very susceptible (likely to be harmed by) to drugs used to kill them. However, today, we find that many pathogens that make people sick are resistant (unlikely to be harmed by) to the same drugs we used to use. Explain, in your own words, where these multi-drug resistant pathogens (superbugs) come from.
