1) Oxygen influence the growth of microbes depends on whether they are obligate aerobes or obligate anaerobes. Various facrtors which influences the growth of these bacteri are: avaialbility of nutrients, temperature; pH, ions and salt concentration and water.
Obligate aerobes depend on oxygen for growth as a terminal electron acceptor in aerobic cellular respiration which is essentially needed for growth. On the other hand, obligate anaerobes rely on fermentation and anaerobic respiration and their enzymes are potential to react with oxidants. Possibility of production of substantial superoxide and hydrogen peroxide due to low-potential flavoproteins for anaerobic respiration.
2) A moistened filter papaer coated with tetramethyl-p-phenylenediamine dihydrochloride is to be smeared by the colony of interest using wooden or platinum loop. Observe for any color change to deep blue or purple within 10-30 seconds.
The tetra-methyl- p-phenylenediamine dihydrochloride is a reagent acts as an artificial electron donor for cytochrome c which upon oxidation gives color change to dark blue or purple compound, indophenol blue.
