the following are the initial steps involved in aseptic transfer to an agar plate. place them In their proper order.(a,b.c,d?)
1. Flame loop
2.insert loop into broth & touch to plate
3. mix tube
a)3,4,2,1
b)3,1,4,2,1
c)1,2,3,4,1
d)4,3,2,1
4) flame tube
b) 3,1,4,2,1
Aseptic techniques are practised by microbiologists for the proper transfer of culture to a medium with out any contamination with any foreign microbes in the environment . the steps involved are,
1st step in transfering a tube culture in the agar plate involves the mixing of tube containg the organism for the proper mixing of organisms in the broth in the tube.
2nd step the sterilization of inoculation loop from base to tip until red hot with flame of bunsen burner
3rd step invove flaming of the tube mouth to sterilize the opening of the tube.
4th step involve inserting the inoculation loop in the broth and then inoculating them into the agar plate with continous stroking of inoculation loop in a zig zag manner in all direction by rotating the plate to ensure that the organisms are evenly spread.
5th step involves the sterilization of the inoculation loop from base to tip tii red hot again with flame of Bunsen burner.
