You want to analyze the potential effects of Cisplatin, a well known chemotherapeutic agent on E-cadherin expression in ASPC1 cells at different concentrations (1, 3 and 10 µM). (15 pts)

a. Name and explain at least 3 techniques you can use for this study in detail.

b. What will be your test conditions for this study?

c. With the below information already given to you, how many microliters of the drug will you add to each condition if you are using a 10 cm dish for cell culture?

- Main stock of Cisplatin is 10 mM

- Test concentrations will be 1, 3 and 10 µM.

Expert Solution

You want to analyze the potential effects of Cisplatin, a well known chemotherapeutic agent on E-cadherin expression in ASPC1 cells at different concentrations (1, 3 and 10 µM). (15 pts)

a. Name and explain at least 3 techniques you can use for this study in detail.

1) measurement of cell viability with flow cytometry

2) measurement of cell viability with membrane integrity

3) measurement of cell viability with colorimetric assay

b. What will be your test conditions for this study?

One control with out the agent, experimental treatments with the three concentrations (1, 3, and 10 uM)

c. With the below information already given to you, how many microliters of the drug will you add to each condition if you are using a 10 cm dish for cell culture?

- Main stock of Cisplatin is 10 mM

- Test concentrations will be 1, 3 and 10 µM.

Initial concentration = 10 mM

final concentration = 0.001 , 0.003 or 0.01 mM

final volume = 10 cm = 392.7 cm2 of area = 39.27 ml

volume needed = Cf * Vf / Ci

For 1 uM

volume needed = 0.001 mM * 39.27 ml / 10 mM

volume needed = 0.0039 ml = 3.9 ul

For 3 uM

volume needed = 0.003 mM * 39.27 ml / 10 mM

volume needed = 0.011 ml = 11 ul

For 10 uM

volume needed = 0.01 mM * 39.27 ml / 10 mM

volume needed = 0.0392 ml = 39.2 ul

You are given data to analyze for a new chemotherapeutic to eliminate malignant tumors and must determine if a significant relationship of any kind exists for the latest drug that was proposed. This new drug, XA98 has shown promising results across multiple repeated cross-sectional studies and now combined evidence was gathered to see if any relationships stand out among the multiple tests that were conducted in the past. The data for XA98 to combat specific cancer tumors can be found...

2 Anisic acid (HC8H7O3) is found naturally in anise, a well-known culinary herb and flavouring agent. It is a monoprotic, weak acid. A 50.0 mL sample of a 0.100 M solution of anisic acid is titrated with 0.165 M potassium hydroxide (KOH). Ka for this acid = 3.39 x 10-5 . 3.2.1 Determine the pH of the anisic acid solution before the titration begins. [4] 3.2.2 What is the pH at the equivalence point? [8]

The mechanism for (possible) trade is that each agent announces a price, and if the buyer's announcement is larger than the seller's announcement, the object is sold to the buyer at the seller's announced price. The Brain Game The purpose of this assignment is to introduce you to the well-known scientific fact that humans do not make decisions rationally. Most of the time, decisions are made without people even realizing the “why” behind their decisions. Our decisions are often driven by our own biases instead of logic or good judgement. These numerous “poor instincts” are called cognitive biases. It is important to identify cognitive biases in Personal Finance because they cause so many of the...

During a board meeting the need to expand internationally was debated although the limited avenues to raise equity and debt domestically may jeopardise such expansion ambitions. One of the choices is to try to raise funds internationally and one of the options would be to list the company on the American market. After talks with the advisory service of...
