ANS- ELISA TEST-->Enzyme-linked
immunosorbent assay is used for the detection of HIV virus , by
measuring the concentration of an analyte (antibodies or antigens)
in the sample solution.
PRINCIPLE--->It is based on the
Antige-antibody complex, which is detected by chromogenic detection
using the enzyme conjugated antibody.
PROCEDURE-
- Antigen/ sample solution is added on a plate.
- Addition of blocking buffer to block remaining protein-binding
sites.
- Specific antibody is added to the coresponds of HIV virus.
- Suitable secondary antibody(HRPO conjugte) is also added on a
plate,that recognises the primary antibody and binds to it.
- Then TMB (Tetramethylbenzidine )substrate is added which is
converted into dectacable (produce colour) form by the help of
HRPO(Horseradish peroxidase) enzyme.
RESULT-
- Light absorbing reader is used for capturing the light released
from the sample, if result is positive dark spot is appeared on a
plate but if it is negative the plate remains same as
previous.
