ANS- ELISA TEST-->Enzyme-linked immunosorbent assay is used for the detection of HIV virus , by measuring the concentration of an analyte (antibodies or antigens) in the sample solution.

PRINCIPLE--->It is based on the Antige-antibody complex, which is detected by chromogenic detection using the enzyme conjugated antibody.

PROCEDURE-

  1. Antigen/ sample solution is added on a plate.
  2. Addition of blocking buffer to block remaining protein-binding sites.
  3. Specific antibody is added to the coresponds of HIV virus.
  4. Suitable secondary antibody(HRPO conjugte) is also added on a plate,that recognises the primary antibody and binds to it.
  5. Then TMB (Tetramethylbenzidine )substrate is added which is converted into dectacable (produce colour) form by the help of HRPO(Horseradish peroxidase) enzyme.

RESULT-

  • Light absorbing reader is used for capturing the light released from the sample, if result is positive dark spot is appeared on a plate but if it is negative the plate remains same as previous.
