Which item is not required for DNA sequencing?

a.

DNA probe

b.

dNTPs

c.

gel electrophoresis

d.

ddNTPs

e.

DNA primer

Solutions

Expert Solution

option a-DNA probe

Requirements for DNA sequencing are-

1.DNA primer-which is Complementary to Known Region to start

2.DNA polymerase

3.4 dNTPs

4.4 ddNTPs

In Sanger sequencing, The Nucleotides are finally loaded into 4 lanes of a single gel and they are separated using gel electrophoresis,

Electrophoresis methods like gel electrophoresis is a method used to seperate different DNA molecules based on their Charge.

DNA is a negatively charged molecule.

In Electrophoresis ...DNA Samples are placed at one end (Near -ve Electrode)and Electric current is Applied .So the negatively charge DNA molecules get moved from the area near -ve Electrode to area near Positive Electrode.

But this movement depends on the Size of the DNA molecules.Smaller DNA moves more and reaches more close towards the Positive electrode. While Large DNA molecules move less.

** This is used to seperate DNA molecules based on their size
