have a bottle of 1 mL of solution of unknown concentration. I have to make the following dilution series: 1:100, 1:200, 1:300, 1:350, 1:400, 1:450, 1:500, 1:550, 1:600. I know I could do 1 microL of solution + 99 microL of buffer to make 1:100 but I would rather do a dilution series. each tube must end with at least 800mircoL. Please explain how I could achieve this, thanks!

Ans: For making dilulution,

We will use dilution farmula C1V1 = C2 V2

C1= Concentration of stock solution

V1= Volume required from stock solution for the dilution

C2= Required concentration

V2= Required volume of diluted solution

Suppose concentration of stock sample is = 1

Required volume with diluted concentartion = 800 uL

1) For 1:100

1 * V1 = 1/100 * 800

V1= 8uL

Means you will need 8ul from stock solution and add into 792 ul buffer for making 800 ul of diluted soution with concentartion 1:100

2) For 1:200

1 * V1 = 1/200 * 800

V1= 4uL

Means you will need 4ul from stock solution and add into 796 ul buffer for making 800 ul of diluted soution with concentration1:200

3) For 1:300

1 * V1 = 1/300 * 800

V1= 2.66uL

Means you will need 2.66ul from stock solution and add into 797.34ul buffer for making 800 ul of diluted soution with concentration1:300

4)  For 1:350

1 * V1 = 1/350 * 800

V1= 2.28uL

Means you will need 2.28ul from stock solution and add into 797.72 ul buffer for making 800 ul of diluted soution with concentration1:350

5) For 1:400

1 * V1 = 1/400 * 800

V1= 2uL

Means you will need 2ul from stock solution and add into 798 ul buffer for making 800 ul of diluted soution with concentration1:400

6) For 1:450

1 * V1 = 1/450 * 800

V1= 1.77uL

Means you will need 1.77ul from stock solution and add into 798.23 ul buffer for making 800 ul of diluted soution with concentration1:450

7) For 1:500

1 * V1 = 1/500* 800

V1= 1.6uL

Means you will need 1.6ul from stock solution and add into 798.4 ul buffer for making 800 ul of diluted soution with concentration1:500

2) For 1:550

1 * V1 = 1/550 * 800

V1= 1.45 uL

Means you will need 1.45ul from stock solution and add into 798.55 ul buffer for making 800 ul of diluted soution with concentration1:550

2) For 1:600

1 * V1 = 1/600 * 800

V1= 1.33uL

Means you will need 1.33ul from stock solution and add into 798.67 ul buffer for making 800 ul of diluted soution with concentration1:600
