Please list and explain minimum 3 techniques used for gene expression analysis on mRNA level??
We can analyse the function or expression of any particular gene segment on mRNA level by these techniques-
(i) Deleting the gene segment: It is a very effective method, if we can delete the gene segment of interest then it is obvious that if the deleted part is responsible for that mRNA expression then the mRNA will not be expressed but if the deleted portion was not responsible then the mRNA expression will be found.
(ii) Changing the genomic sequence or mutation: In this method the gene segment of interest sequence is altered or changed or mutation is done and if the mRNA sequence is also changed or mutated then it is obvious that the gene was responsible for that mRNA expression but if there is no change or mutation is seen in the mRNA sequence then it is obvious that the gene was not responsible for mRNA expression.
(iii) Inserting the gene of interest: In this method extra copies of the gene of interest is added and if the gene is responsible for that mRNA expression then the mRNA will be present more in number but if the gene was not responsible then no change will observe in the mRNA expression level.
