You are preparing two liters of media for pouring LB/Carb agar
plates. The final concentration of
carbenicillin needs to be 100 μg/ml, and you have a stock
carbenicillin solution that is 100
mg/ml. How much of the stock Carb solution do you need to add to
the media after in cools
following autoclaving?
we use the formula C1V1 = C2V2
Where
C1 = concentration of stock solution
V1 = volume of the stock needed
C2 = concentration of working solution
V2 = total volume of solution prepared
So, C1 = 100mg/ml or 100*10^3 microgram/ml
V1 = x
C2 = 100 microgram/ml
V2 = 2*10^3 ml or 2 lt
By using the formula
C1V1 = C2V2
100*10^3 * x = 100 * 2* 10^3
x = 2ml
We need 2 ml of stock solution i,e carbenicillin (100mg/ml) in 2lt of lb-agar.
So, after autoclaving when the media cools down (not solidify) add 2ml of carbenicillin stock solution in it, mix well then pour this to make lb/carb agar plates.
