a)Suspension tests:Here the bacterial culture is placed in the disinfectant solution.After the optimum exposure the culture is looked for the bacteria,if the organism is dies the disinfectant is efficient.

b)Rideal Walker method: it is a test used to estimate the phenol coefficient of the disinfectant.

phenol coefficient is used to compare teh disinfectant with the phenol as the standard effective solution.

Phenol is diluted from 1:400 to 1:800 and the disinfectant is diluted from 1:95 to 1:115. bacterial killing activity is determined by using  S. typhi suspension.

cultures are performed from both the test and phenol at time intervals of 2.5, 5, 7.5 and 10 minutes. The cultu are incubated for 48-72 hours at 37°C. The dilution of disinfectant which disinfects the suspension in a given time is divided by that dilution of phenol which disinfects the suspension in same time gives phenol coefficient of teh disinfectant.

for example

after 5minut the test organism was killed by the test disinfectant at a dilution of 1;600. In the same period the test organism was killed by phenol at a dilution of 1:100

phenol coefficient =600/100=6

effectivity of this disinfectant is greater than the phenol
