  • 5x taq buffer - Resists pH changes by chemically neutralizing small amounts of added acidic or basic compounds, thus maintaining the overall pH of a medium. A taq buffer consists of Tris-HCl, KCl, and MgCl2 with a pH 8.3 at 25 deg Celsius.
  • F and R primers - Forward and reverse primers are used to start the process of replication. These primers are designed to be complementary to the nucleotide sequences at the beginning and the end of the section of DNA that needs to be amplified.
  • Bivalent cations - typically magnesium (Mg) or manganese (Mn) ions; Mg2+. Polymerases require the presence of magnesium to act as a co-factor during the reaction process.
  • deoxynucleoside triphosphates or dNTPs - the building blocks from which the DNA polymerase synthesizes a new DNA strand.
  • DNA polymerase - enzyme that polymerizes new DNA strands.
  • Water - sterile water is used in order to dilute the concentration of the reagents to the proper final concentration.
  • DNA template - contains the DNA target region to amplify.
