Why should a concentration curve be made at the max absorbence wavelength for a given compound?

Solutions

Expert Solution

A concentration curve is made to understand the mode of absorbance of incident beam of light and the pattern of this curve is understood to investigate the properties of the substance. It is important to note here that although mixing of the substance takes place gradually, the absorbance too undergoes changes gradually and maximum absorbance can be found at the maximum solubility of the substance in the medium/solvent. This concentration curve thus must be made at the maximum absorbance of the compound under investigation because of following reasons:

  1. The maximum concentration represents the maximum solubility of the substance in the solvent
  2. The substance remains most stable when it is under maximum solubility and concentration in the solvent, hence, the true optical properties are investigated under this state.
  3. The peak of each substance is different in nature and represents a chemical signature for it, thus helping in identifying the substance.

This is why chemists and bio-chemists often made these observations and calculations at maximum concentration.

In an experiment, why would the concentration of a sample decrease when changing the wavelength for a spectrophotometer from 235 nm to 280 nm? This is just a general question, I would like to know why the absorbance peak would be higher at a wavelength of 235nm rather than at 280nm. Is this normal? or could it an experimetnal error?
