Suppose you have nanometer-sized features in your sample, and would like to resolve the shape as good as you can. Which setting(s) of a Köhler microscope will give you the highest chance of resolving these features and their shape?
By adjusting the following settings on Kohlers microscope we will be able to resolve the features and shape of the nanometer sized materials in the sample:
Realignment procedure steps: These tests helps in making sure that optical components are in focus at the two sets of conjugate image planes; the light source image planes and the specimen image planes.
Once the sample is ready, place it under the microscope. Bring it into focus by moving the specimen prepared. Close the field diaphragm partially closed and the edges of the diaphragm should be in the same conjugate image planes as the specimen, therefore should appear in focus. The field diaphragm is reopened to adjust beyond the field of view.
The condenser lenes and diaphragm can be moved up and down to bring the specimen and it's features in better focus.
