MacConkey agar (MAC):

  • MacConkey agar is a selective (for gram negative) and differential (for lactose fermentation) media.
  • MacConkey agar media inhibits the growth of gram positive media and is used to differentiate gram negative bacteria (example enteric bacteria that can ferment lactose sugar).
  • The inclusion of bile salts and crystal violet dye makes MacConkey agar unsuitable for the growth of gram positive bacteria.
  • Therefore, MacConkey agar media is used for the differentiation of gram negative bacteria that can ferment lactose.
  • The color of colonies will be pink or red for bacteria which can ferment lactose sugar whereas bacteria that can ferment lactose will change the color to bright pink or red.

Eosine Methylene Blue (EMB) media:

  • EMB media is a selective and differential media for coliforms (gram negative enteric bacteria).It is used for the differentiation of lactose fermenting and non-lactose fermenting enteric bacilli.
  • Appearance of color for EMB media:
    • Coliform bacteria; Purplish black colonies
    • Non Coliform Bacteria: Colorless colonies.
