Enzymes can function as labels as their catalytic
properties allow the detection and quantitation of extremely small quantities of immune reactants.

Possibly one of the most popular labels to use in immunoassays is enzymes.These enzymes allow for detection often because they produce an observable colours change in the presence of certain reagents. In some cases these enzymes are exposed to reagents which cause them to produce light or Chemiluminescence.
Immunoassays employ a variety of different labels to allow detection of antibodies and antigens. Labels are typically chemical-linked or conjugated to the desired antibody or antigen.

In colorimetric immunoassays, there are three enzymes that are most frequently used. They are horseradish peroxidase (HRP), alkaline phosphatase (ALP), and β-galactosidase, until now. The detection principle is related to the oxidation or reduction of a substrate under the catalysis of a natural enzyme
