Beer-Lambert law relates the attenuation of light to the properties of the samples in UV-Vis spectrometer; however, in general, the experimental results do not follow the Beer’s law. What are the limitations of Beer’s law?

Solutions

Expert Solution

According to beer lamberts law

Absorbance

is  - molar absorption coefficient C is the cincentration and l is the length of light path

Absorbance is directly preption to concentration.

A can also written as

hence this also can written as

The limitations of this law is

1) This is law is derived for dilute solutions. At higher concentration molecular interaction can occour and deviations can happen.

2) effect of scattering of the light from samples is not accounted.

3) Fluoresent and phosphoresent samples makes deviation from this law.

4) source should be monochromatic.

5) Effect of refractive index of light absorption was not inclued.

Use the following information and the Beer-Lambert law to determine the mass of the lycopene extracted and the mass of lycopene per 100 grams of tomato paste. 2mL of lycopene was extracted from 8 grams of tomato pase. This was then diluted to 5mL. 1ml of the diluted solution was withdrawn and then further diluted to 9mL. The UV-vis absorbance at 502nm was determined to be 0.162. The cell path was 1cm.

Use the Beer-Lambert law to explain how a colorimetric assay works in the spectrophotometer. In your answer, define the following terms; chromophore, extinction coefficient, absorbance, transmission and reagent blank. Most of the spectrophotometers in the teaching lab emit light of wavelengths between 400-700nm. However some spectrophotometers also emit light between 200-400nm. Why is this extended range useful? Provide a specific example. How can you use a spectrophotometer to perform a qualitative analysis of a substance?
