explain what facilitates the renaturation of partially denatured proteins.
Answere:
Denaturation is a process of loss of 3-D conformation of protein to cause a loss of its function. Denatured protein exist in a set of partially folded protein. Denaturation occur :
1. At high temperature 2. Extreme in pH 3. In the presence of detergents, alcohol, acetone, urea, guanidine hydrochloride.
Renaturation is a process of denatured protein can be reversed to its native structure. The native structure of protein will be regained into a stable state and biological activity will return. Most of the times, proteins can be renatured simply by reversing the process of denaturation. Factors facilitating renaturation of proteins are :
I. Temprature : Denatured proteins can be renatured by gradually decreasing temperature of solution. If temperature is reduced too fast, the protein may be trapped in a local minimum rather than reaching native state.
II. pH : Denatured proteins can be renatured by re-adjusting the pH to optimum level i.e. approximately near 7.
III. Removel of denaturing agent : When denaturant is removed from partially denatured protein solution then protein may retain its native structure slowly.
