Researchers investigating cave fish found a new population in a previously undiscovered cave. These fish also lacked eyes, but different from the other populations, their eye development appeared to fail during lens induction. The researchers observed a failure to induce a lens placode. Their next goal was to determine the mechanism that caused loss of eyes in this population.

a) (2 points) The researchers hypothesized that a loss of function mutation in one or more of six possible genes was likely the cause. List the six most likely candidate genes.

b) (3 pts) How could the researchers determine if one of those six genes had a mutation.

c) (5 pts) Assume that the researchers found a mutation in one of those six genes. Describe an experiment the researchers could conduct using the surface fish as a model to determine if the identified mutation was sufficient to cause a failure of eye formation. List the possible results and highlight/underline the result that supports their hypothesis.

Solutions

Expert Solution

1.The below mentioned genes form the genetic basis for eye regression in cave fish.

Cryaa, pitx3,rx3, olfm2a, olfml2a, BCoR.

2. The researchers can do it by sequencing the corresponding gene . Let us suppose it to be Cryaa.

Procedure

Isolate DNA from the cave fish.

Design primers for Cryaa gene.

Amplify the gene by PCR.

Now do Sanger sequencing. For Sanger sequencing take 1micro litre of amplified gene, add only one primer , dNTPS,ddntps, polymerase enzyme, sequencing dye and buffer. Then do the sequencing PCR and proceed to Sanger sequencing. Now analyse the chromatogram of the gene obtained. Compare the sequence to Cryaa gene sequence of surface fish that don't has mutation.

3. For this the gene with mutation has to be incorporated to surface fish. The supposed mutation in Cryaa gene can be made in surface fish by using CRISPR technology.CRISPR helpsin editing gene sequence and thus Cryaa mutation can be made in surface fish in the early embryonic stage and further development of eyes in surface fish can be analysed.

Researchers used a standard fish seine to sample the fish population of Mason Pond. Most of these fish were bluegill (Lepomis macrochirus). Researchers removed fish from the seine and measured these fish on a fish board the boat. Researchers measured each fish to the nearest millimeter. Then, researchers measured all of the fish brought in by your seine, except for fish less than 15 mm.    Size Category Observed Expected 15-35 mm 22 7.33 >35-55 mm 16 5.33 >55 mm...

In zebra fish, assume that a population is found that is polymorphic for a black spot at the base of the dorsal fin. Crosses determine that this is dominant to having no spot. In this population, 36% of all zebra fish have black fin spots. What is the frequency of the recessive allele?

Researchers at Consumer Reports recently found that fish are often mislabeled in grocery stores and restaurants. They are interested to know, however, if the proportion of mislabeling varies by type of fish. They collected data on 400 packages of tuna and 300 packages of mahi mahi and found that 110 and 95 were mislabeled, respectively. a. What are the point estimates for the proportion of tuna and mahi mahi that are mislabeled? b. Provide a 95% confidence interval estimate of...

Researchers investigating temperament in dogs have a database of reactivity scores from a population of N = 1000 puppies.  The reactivity scores are normally distributed with a mean of mu = 175 and a standard deviation of sigma = 20; higher scores indicate higher reactivity (that is, the puppy is more likely to become aroused by novel stimuli). Dr. Kobe is also conducting temperament testing, but his local shelter is much smaller than Dr. Santos’.  Thus, he is only able to obtain...

Researchers are investigating how eye color affects trustworthiness. In their experiment, the researchers took photographs of 80 students, 40 of whom had brown eyes, and 40 of whom had blue eyes. They then recruited 105 participants, who judged each of the photographs on a 10 points trustworthiness scale (1pt=very untrustworthy, 10pts=very trustworthy). The mean score for brown-eyed students was 5.55, with a standard deviation of 1.68. For blue-eyed students the mean score was 4.62, with a standard deviation of 1.53....

Fish Ventilation Most fish use gills for respiration in water and researchers can observe how fast a fish’s gill cover beats to study ventilation, much like we might observe breathing rate for a person. Professor Brad Baldwin is interested in how water chemistry might affect gill beat rates. In one experiment he randomly assigned fish to tanks with different levels of calcium. One tank was low in calcium (0.71 mg/L), the second tank had a medium amount (5.24 mg/L), and...

Researchers are investigating the effect of storage temperature on bacterial growth for two types of seafood. They set up the experiment to evaluate 3 storage temperatures. There were 9 storage units that were available, and so they randomly selected 3 storage units to be used for each storage temperature, and both seafood types were stored in each unit. After 2 weeks, bacterial counts were made. After taking a logarithmic transformation of the counts, they produced the following ANOVA: Type 3...

You are reading a research study where the researchers are investigating osteoarthritis pain. In the study, subjects are given either an anti-inflammatory drug injection or a saline injection to reduce osteoarthritis joint pain. A study subject reported that since she received her study injection, her arthritis pain does not seem to have improved. In fact, her pain is getting worse. What is a possible reason for her continued osteoarthritis pain? The research subject likely received the anti-inflammatory study drug, but...
